1/6 We use the chemical Mandipropamid (short: Mandi) to selectively control protein interactions in cellulo and in vivo. The Mandi system is based on plant proteins which means it doesn’t cause off-target effects in mammalian cells.
2/6 In contrast to other plant-based CIPP systems, Mandi is highly cell permeable. We used it to control intracellular protein-protein interactions with high efficiency and response times of less than two seconds.
3/6 We also show that Mandi can be used for in vivo control of protein interactions making it a great tool for use in developmental biology, complex cell culture systems or organoid model systems.
4/6 Combining Mandi with another plant-based CIPP system allowed us to reversibly shuttle proteins between subcellular targets. This will allow complex logic gating based on small molecules in the future.
5/6 Finally, we used nanobodies to target the Mandi system to endogenous, untagged targets. We believe that this might be an effective solution to control protein interactions and localization where tagging of targets is not possible.
