If you don’t know the play, Viola and Sebastian are twins and Viola is supposed to look exactly like Sebastian when she disguises herself as Cesario, so it would be a fun riff on Walter and Gary being brothers in 2011 Muppets film.
Oscar Isaac would be the only other major human player as Duke Orsino and Miss Piggy would be Countess Olivia.
So Miss Piggy would then spend the entire movie trying to mack on Anne Hathaway, only to turn around and think that Kermit is the guy she’s been trying to kiss and marry for the whole time.
Meanwhile, Sam the Eagle is Benvolio and he’s getting absolutely trolled by Sir Toby Belch (Rizzo), Sir Andrew Augecheek (Gonzo), Feste (Fozzy the Bear), Maria (Yolanda), and Fabian (Pepe the Prawn).
Have Kevin Bishop (who played Jim Hawkins in Muppet Treasure Island) as the sea captain Antonio who rescues Sebastian and Sweetums as the first captain that rescues Viola.

Unfortunately Twelfth Night doesn’t present itself to opportunities to include Doctor Teeth and the Electric Mayhem the way Much Ado does, but they’ll find a way. They always do.
I don’t know if Oscar Isaac has done Shakespeare, but I feel like he can be appropriately melodramatic for both the original text AND trying to court a pig.
