Personal take, but for folks applying for jobs (in comms/gaming): I read EVERY cover letter.

Know the problems/opportunity I want to solve (which a good job desc. should have), then connect your experiences/insights to my problem.
BELIEVE IN YOURSELF!
Tell me why I need you! Experience is great, but EXP is just shorthand for, "I've solved problems like yours before."

If you think I should hire you over everyone, tell me! Even if your experience is ~weird~. Help me think outside the box as to who can or should solve this problem (i.e. you). I need a fresh perspective on this challenge. I want to be surprised (pleasantly) by the solution!
If you have 'standard' experience and no cover letter, I may pass you up if I've found sharper candidates that day.
